Fastest air route

I’d like to know the fastest air route at a given altitude between two points. Is this possible?

Shortest flight route on Earth is always an arc of a great circle (aka. geodesic line), as explained here or there. Depending on aircraft speed, shortest path would always get the fastest route. The same for boats. And of course winds or ocean streams could change a lot this pure geometrical computation...

I guess you mean the fastest air route in term of time?
In term of distance, Windy shows it. At starting point click your right mouse button to select Distance & planning. It gives the shortest distance to the end point (according to an arc of great circle amsl).
In term of time, I imagine you want to know the influence of wind at different altitudes.
Have you tried the Route planner or the Flight planner (plugin)? They calculate the duration of the flight, but at a fixed speed. However they give the wind speed and direction at different altitude along your flight route. Aviation specialists may give you a more precise answer, as @rittels.
