In Distance and Planning, Is it possible to insert a layover at a given waypoint - for example, start now at waypoint 1, go to 2, 3, 4, then at waypoint 5 stop for 12 hours, then go to 6, 7, 8?

No, that's not possible.But you can plan two routes.
One to the stopover (WP 1, 2, 3, 4, 5), the other beyond (WP 6, 7, 8) since you can set the departure time as you wish.
